Joint elimination and joint transduction of the determinants of penicillinase production and resistance to macrolide antibiotics. J. Bacteriol. 89:988-992. 1965.-Strains of Staphylococcus aureus, which show high resistance to macrolide antibiotics (erythromycin, oleandomycin, leucomycin, and spiramycin) and the capacity to produce penicillinase, have been isolated from clinical sources. The determinants of penicillinase production (PCase(+)) and resistance to macrolide antibiotics (MAC(r)) of these strains were irreversibly eliminated by treatment with acridine or with ultraviolet light. Among the 18 strains tested, PCase(+) and MAC(r) were eliminated from all strains except one, which lost only PCase(+) but not MAC(r). The characters PCase(+) and MAC(r) were jointly transduced with the aid of phage lysates, obtained from the resistant donors by ultraviolet irradiation, into staphylococcal strains sensitive to PC and MAC. Segregation of PCase(+) and MAC(r) was rarely observed after transduction. From these results, it is suggested that the determinants of both PCase(+) and MAC(r) of staphylococci are located close together on a single genetic element, i.e., a plasmid (or episome), which exists extrachromosomally.

Transduction and elimination of resistance determinants in methicillin-resistant Staphylococcus aureus.
Elimination and transduction of drug resistance was examined in methicillin-resistant strains of Staphylococcus aureus. Irreversible spontaneous loss and "curing" by aging of cultures and by treatment with ethidium bromide indicated that the determinants for penicillinase production and chloramphenicol resistance, and probably also for neomycin resistance, were located extrachromosomally. On th...

Background: Antibiotics called macrolide, lincosamide and streptogramin B (MLSB) are being used to treat staphylococci infections. Multiple pathways that impart resistance to MLSB antibiotics have been confirmed to cause clinical failure. The present work aimed to determine the frequency of constitutive and inducible clindamycin resistant among coagulase-negative staphylococci (CoNS) isolates o...
متن کاملLability of Resistance to Aminoglycoside Antibiotics in Pseudomonas Aeruginosa
The determinants for resistance to aminoglycoside antibiotics in some strains of Pseudomonas aeruginosa were irreversively lost during storage or by treatment with ethidium bromide. This fact has indicated the presence of plasmids governing resistance to aminoglycosides in the pseudomonads as well as R factor in gram-negative enteric bacteria and plasmid in staphylococci. Many papers have descr...
متن کاملArtificial elimination of drug resistance from group A beta-hemolytic streptococci.
Ten strains of beta-hemolytic streptococci were tested for elimination of resistance to macrolide antibiotics, tetracycline, and chloramphenicol. Six of the strains lost resistance after cultivation at 41 C or addition of acriflavine (0.2 mug/ml). These results suggest that determinants governing resistance to these antibiotics are located on extrachromosomal genetic elements (plasmids) that ar...
